Reconstitution practices affect downstream measurements. The dry powder is typically dissolved in sterile water or a suitable aqueous buffer, then mixed gently rather than vortexed at high speed. Visible particles or cloudiness suggest incomplete dissolution or contamination and should be investigated. For long-term storage, aliquots should be labeled with concentration, solvent, and date. Open questions include how different buffers alter peptide conformation and whether specific container materials adsorb the peptide. Those variables can change apparent concentration in assays even when the chemical identity is correct.
Lyophilized TB-500 is hygroscopic and should be kept dry before use. The usual storage recommendation for the solid is -20 °C, protected from light and moisture. Once dissolved, the peptide is less stable, and repeated freeze-thaw cycles can promote aggregation or degradation. Laboratories often divide a reconstituted solution into single-use aliquots and store them at -80 °C. Exact stability limits depend on buffer, pH, and concentration, so published data do not define a single universal condition.
Thymosin beta-4 contains 43 amino acids and has a reported molecular mass near 4963 Da. The short fragment most often associated with the TB-500 label, an acetylated chain beginning LKKTETQ, has a reported mass near 889 Da, so the two are easily separated in analytical work. Mass spectrometry and amino acid analysis can confirm which material is present in a given sample. Statements treating TB-500 and thymosin beta-4 as interchangeable are therefore imprecise, even though the two appear together in much of the same literature.
Interest in the compound comes largely from studies of the parent protein, which participates in actin sequestration, cell migration and tissue repair processes. Whether a short fragment reproduces those activities is a separate question that remains open in the published record. Many summaries describe mechanisms by analogy to thymosin beta-4 rather than from direct measurements on the fragment. Claims about activity should be treated as provisional unless a cited study specifies the exact peptide, its purity and the assay used.

TB-500 is a catalogue name applied to a synthetic peptide related to thymosin beta-4, an actin-binding protein found in most mammalian cells. Suppliers do not use the label consistently: some describe it as the full 43-residue protein, others as a short fragment from the actin-binding region, and others as a related tetrapeptide. Because the name is commercial rather than chemical, two products sold under it may not contain the same molecule. This naming ambiguity is the first point to check in any description of the material.
The most frequently cited identity is a seven-residue fragment with the sequence LKKTETQ, taken from the actin-binding domain of the parent protein. A separate molecule, N-acetyl-seryl-aspartyl-lysyl-proline, often shortened to Ac-SDKP, derives from the same protein's N-terminal region and appears in overlapping literature. Reported molecular masses therefore differ between sources, and a mass value on its own does not establish which fragment is present. Confirmation requires a defined sequence rather than a single number.
Dry powder is commonly held at minus twenty degrees Celsius, with some suppliers recommending lower temperatures for long-term archival storage. Once dissolved, solutions are typically kept cold and protected from light, since aqueous peptide solutions can lose integrity through hydrolysis or oxidation over time. Stability data specific to this fragment are limited in the public literature, and much of the guidance comes from general peptide handling practice rather than from controlled degradation studies. Users therefore treat stated shelf lives as approximate rather than fixed.
Identity and purity are normally assessed with reversed-phase high-performance liquid chromatography, paired with mass spectrometry to confirm molecular mass. A certificate of analysis reports a purity percentage, usually derived from chromatographic peak area, but that figure does not by itself prove a correct sequence or the absence of counterions. Independent verification may include amino acid analysis or peptide mapping. Batch-to-batch variation is a documented concern in the research chemical market, and the gap between a quoted purity value and actual peptide content can be substantial when the material is a salt or retains residual water.
Lyophilized peptide arrives as a dry cake that should stay sealed until use. Reconstitution is generally performed with sterile water or a buffered solution, and the resulting liquid should be handled gently to limit mechanical stress. Repeated freeze-thaw cycles are widely described as harmful to short peptides, so dividing a reconstituted batch into single-use portions is a common practice. Laboratories also record the solvent, concentration, and date of preparation on the vial label to keep later measurements traceable.
Literature and online discussion often conflate TB-500 with full-length thymosin beta-4, even though the two differ in size and are not interchangeable in analytical terms. The fragment is produced by solid-phase peptide synthesis, and the product is a defined seven-residue chain rather than a biological extract. Because the term is a trade-style label, two vendors may supply materials of the same nominal sequence but different counter-ion content, purity, or water content. Comparisons across studies are therefore difficult unless the exact sequence and purity are reported.
TB-500 is a research peptide whose sequence matches residues 17 to 23 of thymosin beta-4, a 43-residue protein present in most mammalian cells. The chain is seven amino acids long, written as LKKTETQ, and is normally supplied with an acetyl group on the N-terminus. Suppliers list it as a lyophilised powder under the code name TB-500, and the same sequence appears elsewhere in catalogues as the thymosin beta-4 actin-binding fragment. The label is commercial rather than systematic, so no single authority fixes exactly what TB-500 denotes.
Thymosin beta-4 was isolated from calf thymus in the early 1980s and later characterised as an abundant intracellular actin-sequestering protein. Interest in short synthetic fragments grew once the actin-binding motif had been mapped to the middle of the sequence. TB-500 came out of that line of work as a truncated analogue rather than a natural isolate, and it is now sold mainly to laboratories. Published studies on the fragment have been largely in vitro or in animal models, and controlled human trials remain sparse, so claims about effects in people rest on extrapolation.
Several names appear in scientific and commercial contexts for this peptide. The label TB-500 is informal and does not follow standard biochemical nomenclature. Research articles more often describe the compound as a thymosin beta-4 fragment, Tβ4 fragment, or by its sequence Ac-LKKTETQ. Confusing TB-500 with full-length thymosin beta-4 can lead to incorrect assumptions about activity because the fragment lacks the remaining residues of the parent protein. The relationship between fragment and parent protein remains an active area of study.
Regulatory status differs by country, but TB-500 is not an approved pharmaceutical in major jurisdictions. It is commonly sold as a research chemical for laboratory use, which places responsibility for identity and purity on the supplier and the laboratory. Published human data are limited, and most reports involve preclinical models or cell culture. Questions about whether the fragment mimics all actions of thymosin beta-4, and under which conditions, remain open. Independent verification of any material is therefore a practical requirement in research settings.

Crucibles used to contain plutonium need to be able to withstand its strongly reducing properties. Refractory metals such as tantalum and tungsten along with the more stable oxides, borides, carbides, nitrides and silicides can tolerate this. Melting in an electric arc furnace can be used to produce small ingots of the metal without the need for a crucible. Cerium is used as a chemical simulant of plutonium for development of containment, extraction, and other technologies.
Explorer-naturalists such as Alexander von Humboldt investigated the interaction between organisms and their environment, and the ways this relationship depends on geography—laying the foundations for biogeography, ecology and ethology. Naturalists began to reject essentialism and consider the importance of extinction and the mutability of species. Cell theory provided a new perspective on the fundamental basis of life. These developments, as well as the results from embryology and paleontology, were synthesized in Charles Darwin's theory of evolution by natural selection. The end of the 19th century saw the fall of spontaneous generation and the rise of the germ theory of disease, though the mechanism of inheritance remained a mystery. In the early 20th century, the rediscovery of Mendel's work in botany by Carl Correns led to the rapid development of genetics applied to fruit flies by Thomas Hunt Morgan and his students, and by the 1930s the combination of population genetics and natural selection in the "neo-Darwinian synthesis". New disciplines developed rapidly, especially after Watson and Crick proposed the structure of DNA. Following the establishment of the Central Dogma and the cracking of the genetic code, biology was largely split between organismal biology—the fields that deal with whole organisms and groups of organisms—and the fields related to cellular and molecular biology.
== Sequence representation == Following the header line, the actual sequence is represented. Sequences may be protein sequences or nucleic acid sequences, and they can contain gaps or alignment characters (see sequence alignment). Sequences are expected to be represented in the standard IUB/IUPAC amino acid and nucleic acid codes, with these exceptions: lower-case letters are accepted and are mapped into upper-case; a single hyphen or dash can be used to represent a gap character; and in amino acid sequences, U and * are acceptable letters (see below). Numerical digits are not allowed but are used in some databases to indicate the position in the sequence. The nucleic acid codes supported are:

==== Below 1% ==== There are few studies of the health effects of long-term continuous CO2 exposure on humans and animals at levels below 1%. Occupational CO2 exposure limits have been set in the United States at 0.5% (5000 ppm) for an eight-hour period. At this CO2 concentration, International Space Station crew experienced headaches, lethargy, mental slowness, emotional irritation, and sleep disruption. Studies in animals at 0.5% CO2 have demonstrated kidney calcification and bone loss after eight weeks of exposure. A study of humans exposed in 2.5 hour sessions demonstrated significant negative effects on cognitive abilities at concentrations as low as 0.1% (1000 ppm) CO2 likely due to CO2 induced increases in cerebral blood flow. Another study observed a decline in basic activity level and information usage at 1000 ppm, when compared to 500 ppm. However a review of the literature found that a reliable subset of studies on the phenomenon of carbon dioxide induced cognitive impairment to only show a small effect on high-level decision making (for concentrations below 5000 ppm). Most of the studies were confounded by inadequate study designs, environmental comfort, uncertainties in exposure doses and differing cognitive assessments used. Similarly a study on the effects of the concentration of CO2 in motorcycle helmets has been criticized for having dubious methodology in not noting the self-reports of motorcycle riders and taking measurements using mannequins.

=== 1974 === 16 April: Thérèse Siegenthaler (21). A sociology student at Greenwich Community College, Siegenthaler was abducted by the Wests as she hitchhiked from South London to Holyhead. Fred mistook her Swiss accent for Dutch and always referred to her as either "the Dutch girl" or "Tulip". She was reported missing to Scotland Yard by her family in Switzerland when communication from their daughter abruptly ceased. Fred later further concealed Siegenthaler's remains by building a false chimney breast on her grave. 15 November: Shirley Hubbard (15). A foster child abducted from a Droitwich bus stop close to the River Severn as she travelled home from a date. Aged 15 when murdered, Hubbard had been attending work experience in Worcester and was last seen by her boyfriend, having promised to meet him the next day. Her dismembered remains were found in a section of the cellar known to the family as the "Marilyn Monroe area". Hubbard's head had been completely covered in tape, with a one-eighth-inch (3.2 mm) diameter rubber tube inserted three inches (7.6 cm) into her nasal cavity to enable her to breathe.

==== Precipitation amount effect ==== In contrast to temperature control at high latitudes, the isotopic composition of precipitation in the tropics is mainly influenced by rainfall amount (negative correlation). This "amount effect" is also observed for summer precipitation in the subtropics. Willi Dansgaard, who first proposed the term "amount effect", suggested several possible reasons for the correlation: (1) As cooling and condensation progress, the rainfall isotopic composition reflects an integrated isotopic depletion by the Rayleigh process; (2) A small amount of rainfall is more likely to be influenced by evaporation and exchange with surrounding moisture, which tend to make it more isotopically enriched. At low latitudes, the amount effect for δ18O is around −1.6‰/100mm precipitation increase at island stations, and −2.0‰/100mm at continental stations. It was also noted that the amount effect was most pronounced when comparing isotopic composition of monthly precipitation at different places in the tropics. The amount effect is also expected for HIC, but there are not as many calibration studies. Across southeast Asia, the δD sensitivity to monthly precipitation amount varies between −15 and −25‰/100mm depending on location. In temperate regions, the isotopic composition of precipitation is dominated by rainfall amount in summer, but more controlled by temperature in the winter. The amount effect may also be complicated by changes in regional moisture sources.
Two approaches to synthesis are used commercially. In chemical synthesis, aspartame is synthesized using the L enantiomer of phenylalanine. The two carboxyl groups of aspartic acid are joined into an anhydride, and the amino group is protected with a formyl group as the formamide, by treatment of aspartic acid with a mixture of formic acid and acetic anhydride. Phenylalanine is converted to its methyl ester and combined with the N-formyl aspartic anhydride; then the protecting group is removed from aspartic nitrogen by acid hydrolysis. The drawback of this technique is that a byproduct, the bitter-tasting β-form, is produced when the wrong carboxyl group from aspartic acid anhydride links to phenylalanine, with desired and undesired isomer forming in a 4:1 ratio. Another process used to synthesize aspartame is to use an enzyme from Bacillus thermoproteolyticus to catalyze the condensation of chemically altered amino acids. This will produce high yields of aspartame without the β-form byproduct. A variant of this method, which has not been used commercially, uses unmodified aspartic acid but produces low yields. Methods for directly producing aspartyl-phenylalanine by enzymatic means, followed by chemical methylation, have also been tried but not scaled for industrial production.
UHDTV-1 is 3840 pixels wide by 2160 pixels tall (8.3 megapixels), which is four times as many pixels as the 1920 × 1080 (2.07 megapixels) of 1080p HDTV (full HDTV). Also known as 2160p, and 4K UHD. Although roughly similar in resolution to 4K digital cinema formats, it should not be confused with other 4K resolutions such as the 4096 × 2160 DCI 4K/Cinema 4K. The total number of pixels of RGB stripe type is 8.3 megapixels. UHDTV-2 is 7680 pixels wide by 4320 pixels tall (33.18 megapixels), also referred to as 4320p and 8K UHD, which is sixteen times as many pixels as 1080p HDTV, which brings it closer to the detail level of 15/70 mm IMAX. NHK advertises the 8K UHDTV format with 22.2 surround sound as Super Hi-Vision, which can be broadcast with H.264 codecs.
== Interactions == Ramelteon has been evaluated for potential drug interactions with the following medications and showed no significant effects: omeprazole, theophylline, dextromethorphan, and midazolam, digoxin and warfarin. There were no clinically meaningful effects when ramelteon was coadministered with any of these drugs. A drug interaction study showed that there were no clinically meaningful effects or an increase in adverse events when ramelteon and the SSRI Prozac (fluoxetine) were coadministered. When coadministered with ramelteon, fluvoxamine (strong CYP1A2 inhibitor) increased AUC approximately 190-fold, and the Cmax increased approximately 70-fold, compared to ramelteon administered alone. Ramelteon and fluvoxamine should not be coadministered. Ramelteon has significant drug–drug interaction with the following drugs: amiodarone, ciprofloxacin, fluvoxamine, ticlopidine. Ramelteon should be administered with caution in patients taking other CYP1A2 inhibitors, strong CYP3A4 inhibitors such as ketoconazole, and strong CYP2C9 inhibitors such as fluconazole. Efficacy may be reduced when ramelteon is used in combination with potent CYP enzyme inducers such as rifampin, since ramelteon concentrations may be decreased.

Reverse-phase HPLC assesses purity, and mass spectrometry confirms molecular mass. The combination helps distinguish the target peptide from truncated or modified impurities.
Buffer composition, pH, adsorption to containers, and freeze-thaw history can all affect the amount of intact peptide in solution. These factors may change results even when the starting material is chemically correct.
No. TB-500 is a trade-style label used for a synthetic peptide described as a fragment of thymosin beta-4, while thymosin beta-4 is the full 43-residue protein. The two differ in size and are not interchangeable terms in analytical work.
